Agonist Efficiency Estimated from Concentration Response Curve

چکیده
Concentration-response curves (CRCs) are characterized by a midpoint (EC50) and high-concentration asymptote (POmax) that relate to the agonist's affinity efficacy. Recently, third agonist attribute, efficiency, was defined as fraction of ligand binding energy is applied conformational change activates receptor. We show efficiency can be calculated from EC50 POmax estimate its value for 17 agonists nicotinic acetylcholine receptor (AChR). Adult skeletal muscle AChRs were expressed in HEK cells CRCs compiled single-channel currents POmax. There two populations having an 52±2% (n=11) or 41±3% (n=6) (mean±s.d). Agonists with smaller head-group volume (70±9 A3) have higher than those larger (102±18 A3). Examples high-efficiency/small-volume group include quaternary amines (ACh, carbamylcholine, tetramethyammonium others) well secondary (nornicotine, anabaesine DMPP). Low-efficiency/large-volume smoking-cessation drugs (varenicline cytosine), cations (tetraethlyammonium tetramethylphosphonium) (epibatidine, anatoxin). αY190A only one 22 site mutations affect ACh, switching it higher- lower-efficiency population. If known, estimated receptor's constitutive level activity any CRC.

Chlorophyll Concentration Estimated from Irradiance Measurements at Fluctuating Depths
Variations in the spectral shape of underwater irradiance at a fixed depth can be used to describe changes in water column constituents, such as chlorophyll concentration. However, random fluctuations of the depth of the monitoring sensor, caused by current variations, makes interpretation more challenging. Even in homogeneous water, the spectral underwater light field varies with depth because...
متن کاملBootstrap estimated true and false positive rates and ROC curve
Diagnostic studies and new biomarkers are assessed by the estimated true and false positive rates of the classification rule. One diagnostic rule is considered for highdimensional predictor data. Cross-validation and the leave-one-out bootstrap are discussed to estimate true and false positive rates of classifiers by the machine learning methods Adaboost, Bagging, Random Forest, (penalized) log...
متن کاملDetection limit estimated from slope of calibration curve: an application to competitive ELISA.
This paper theoretically derives a general rule that while the slope of the semi-logarithmic plot (Y vs. log X) of a calibration curve varies depending on analyte concentration, X, the slope takes a specific value at the detection limit (L(D)). This rule holds good irrespective of the shape of the calibration curve (linear or non-linear) and in this paper, is applied to competitive ELISA (enzym...
متن کاملBetter Efficiency Curve - let Based Image Fusion
In computer vision, Multi-sensor Image fusion is the process of combining relevant information from two or more images into a single image. The resulting image will be more informative than any of the input images. In remote sensing applications, the increasing availability of space borne sensors gives a motivation for different image fusion algorithms. Most of the available equipment is not ca...
متن کاملStatistical efficiency of curve fitting algorithms
We study the problem of fitting parametrized curves to noisy data. Under certain assumptions (known as Cartesian and radial functional models), we derive asymptotic expressions for the bias and the covariance matrix of the parameter estimates. We also extend Kanatani’s version of the Cramer-Rao lower bound, which he proved for unbiased estimates only, to more general estimates that include many...
